- Sandugo is the story of two brothers who grew up separately and ended up on opposing sides. JC and Aris are fraternal twins who did things together as kids. But while Aris is a strong and athletic boy, JC is suffering from a congenital heart ailment and is in dire need of an operation. Their parents tried their best to earn the money for the treatment but JC's time is running out. In an act of desperation, their mother gives up Aris for adoption, receiving a hefty sum in return. Soon, JC and Leo will face each other from opposing sides of the law not knowing they are brothers. Will they wage war against each other or will their hearts remember that they are brothers of one blood.
- Elton Chiong (Jay Manalo) is a businessman who has three wives. Chona (Zsa Zsa Padilla), a pure Chinese, is his legal wife and has three children: Hamilton (John Prats), a homosexual; Hibiscus (Julianne Lee); and Anthurium (Ella Guevara). Patty (Chey Pie Picache), Chinese with a Visayan accent, is Elton's good-luck charm and the mother of his son Nixon (JC De Vera). Filipina Gloria (Rufa Mae Quinto) is Elton's mistress.

- Marge, a single mom and interior designer, meets Jules, a widower and small-time contractor, in a small accident that starts off their acquaintance on the wrong foot. Teamed up for a renovation project in the scenic town of Lobo, Batangas, the two develop an unexpected friendship. As they are forced to work closely together, feelings start bubbling under the surface.
- Jules musters up the courage to declare his intentions for Marge as the renovation project nears completion. Poised to make his move, someone important from Marge's past arrives and throws his plans up in the air. Now, Jules must not only vie for Marge's affection, but also prove himself to be the better man.
- Jules keeps getting his attempts at wooing Marge thwarted by Richard. He tries to win some brownie points when he and his daughter JR are invited to a picnic that Richard organized. As they spend time together, several secrets are revealed.
- JR and Charles make their way to a hidden island, where they bond and share how they feel about their parents' romance. Jules, Marge, and Richard rush to get to their children, thinking they have ran away and are in need of rescue. Meanwhile, the source of the bad blood between Elizabeth and Malu is revealed.
- Jules works to win over Elizabeth, and shows that he is capable of providing for Marge and her son. As he tries to impress his girlfriend's family, he unknowingly neglects his friends.
- As Marge turns down a surprise marriage proposal from Richard, Jules is brought to thinking that he is getting in the way of reuniting their family. Meanwhile, things between Elizabeth and Malu come to a head in a catfight.
- Jules decides to step back to keep himself from getting in the way of Marge's chance to put her family back together. He strives to focus on the construction firm, while Marge halfheartedly prepares for her family's flight to London. Torn between what their minds and hearts are saying, their loved ones give them a little nudge to go for what they truly want.
- Two months after the wedding, Jules and Marge settle their blended family in a small cottage near Elizabeth's mansion. But as they navigate their new life and roles, some things remain the same. It gets more complicated with Richard making a decision much to the couple's dismay, and the return of an old friend from Jules' past.
- Jules' childhood friend Mica returns to town and hires the newlyweds to renovate her new home. Marge makes a mistake on her presentation which causes a misunderstanding between her and Jules. An encounter between Richard and Lolo Bruno prompts Charles and Kara to break up.
- Now that the honeymoon period is starting to wear off, Marge and Jules, as well as the rest of the household, are getting on each other's nerves. The family learns to compromise and handle each other's temper in order to have a home sweet home.
- Marge goes increasingly suspicious about the time Jules spends with Mica, and tries to resolve this once and for all. Clueless on why Marge suddenly went cold, Jules decides to reignite the spark and relive their courtship days, with the help of his friends. In an effort to make up with Kara, Charles shows off to her friends, leading to a disaster.
- Marge attempts to have some alone time with Jules, but work and his new client, Mica, seem to always get in the way. Charles helps Kara prepare for Grandpa Bruno's birthday, and Richard sees this as an opportunity to be the 'best dad ever'. With Elizabeth's assistance, he offers to help his son, eventually, getting the whole family involved.
- Marge and Jules find out they are having a baby, but are warned to prepare for some difficulties along the way. Richard announces his role in the town's newest construction company.
- Elizabeth and Malu badger Marge with pregnancy advice. Richard tries to recruit Jules' friends to the new construction company. Kara's old friend Kulas keeps winning everyone over, while Tommy takes a shine to newcomer Agnes.
- Jules and the family plan a gender reveal party for Marge. Charles' insecurities about Kulas come to a head during a beach cleanup. A complication threatens Marge and the baby.
- Rumors of an "aswang" on the loose and an elevated blood pressure set the pregnant Marge on the edge. Charles and JR plan a double date to keep things platonic between Kara and Kulas. Tommy recruits Jules, Bart, and Drew to help him serenade Agnes.
